Estudio serológico y molecular de la enfermedad infecciosa de la bolsa de Fabricio (IBDV) en dos lotes de una progenie de reproductoras pesadas

Resumen
La presente tesis pretendió contribuir al conocimiento de la dinámica de los
anticuerpos matemos en la progenie de dos lotes de reproductoras pesadas ( un lote que se
encontraba al inicio de su ciclo de postura y el otro en la etapa intermedia ), así como al
entendimiento de la relaci6n entre los niveles de anticuerpos, la vacunación y la infección en
condiciones de campo con cepas del virus de la enfermedad infecciosa de la bursa (IBDV),
mediante la identificaci6n molecular de las cepas presentes empleando la técnica de reacción
en cadena de la polimerasa o PCR.
Se obtuvo un total de 252 muestras de suero, provenientes de pollos de engorde de la
empresa Avícola RICURA: granjas Ay B, los cuales se evaluaron mediante la técnica
inmunoenzimática (ELISA) indirecto y neutralizaci6n viral. Además, se colectó 20 muestras
de bursa, que fueron procesadas mediante (PCR).
Los resultados serológicos obtenidos mediante la técnica inmunoenzimática asociada a
enzimas (ELISA) y neutralizaci6n revelaron un 100% de seropositividad para los primeros
días de muestreo (1,7 días) con un descenso gradual y sostenido basta alcanzar una
seronegatividad de 100% al día 28 para las aves de ambas granjas. Para el día 42 se observa un
incremento en los niveles de anticuerpos con ambas técnicas.
El análisis molecular mediante PCR, reveló la presencia del gen VP2 en las muestras
de vacuna, cepa variante y en tres “pooles” de muestras de bursa colectadas en el campo.
The following work attempts to make a contribution to the knowledge of the dynamics of the maternal antibodies in the progeny of two lots of the broilers breeder flocks ( one of the lots was in the beginning of the cycle while the other in the intermediate stage) as in the understanding of the relationship within the levels of the antibodies, the vaccination and the infection in field conditions with IBDV strains, through the molecular identification and characterization strains using Polymerase Chain Reaction (PCR). A total of 252 serum samples were obtained broilers from the bird-raising company RICURA: farm A and B, which were evaluated using indirect ELISA technique and viral neutralization. Besides, 20 bursa samples were collected and processed through the PCR technique. The ELISA and neutralization serologic results revealed a seropositivity of 100% the first days ( day 1 and 7) with a gradual descent until seronegativity of 100% was reached on day 28 for both farms. On day 42 an antibodies increment was observed with both techniques. The PCR molecular analysis revealed the presence of the VP2 gen on the vaccine, variant strain and three pool of bursa samples.
