Resumen
La teca (Tectona grandis L. f.), es un árbol originario del sureste asiático, considerado de gran valor
económico principalmente por su madera. Como parte de los esfuerzos por el mejoramiento genético de
esta especie, se han desarrollado protocolos para la crioconservación de polen, sin embargo, es
indispensable asegurar la viabilidad y germinación eficiente del mismo para futuros cruzamientos de
material seleccionado. El objetivo del presente trabajo fue evaluar el efecto de un análogo espirostánico
de brasinoesteriode (Biobras-16®) y una mezcla de oligogalacturónidos (Pectimorf®) sobre la
germinación in vitro, tamaño del polen y longitud del tubo polínico de polen de teca crioconservado.
Los experimentos consistieron en evaluar la germinación de polen a 0, 4 y 8 meses de crioconservación,
en medio de cultivo sin suplementos (M1), suplementado con 0,001 y 0,050 mg L-1de Biobras-16® y 5
y 10 mg L-1 de Pectimorf®. Los datos de germinación fueron analizados mediante un Modelo Lineal
General, mientras que la longitud del tubo polínico y el área del polen mediante análisis de varianza
(ANOVA) y análisis de correlación. Se demostró que ambos productos bioactivos favorecieron la germinación in vitro de los granos de polen crioconservados durante 4 y 8 meses, donde Biobras-16 a
0,001 mg L-1 mostró resultados más consistentes. Pectimorf estimuló la elongación del tubo polínico,
lo cual podría favorecer los procesos de fecundación.
Teak (Tectona grandis L. f.), a tree native to Southeast Asia, is considered of great economic value mainly for its wood. As part of the efforts for the genetic improvement of this species, protocols have been developed for the cryopreservation of pollen, however, it is essential to ensure the viability and efficient germination of the same for future crosses of selected material. The objective of the present work was to evaluate the effect of a spirostannic analogue of brassinosteriod (Biobras-16) and a mixture of oligogalacturonides (Pectimorph) on in vitro germination, pollen size and pollen tube length of cryopreserved teak pollen. The experiments consisted of evaluating pollen germination at 0, 4 and 8 months of cryopreservation, in unsupplemented culture medium (M1), supplemented with 0.001 and 0.050 mg L-1 of Biobras-16® and 5 and 10 mg L-1 of Pectimorf®. Germination data were analyzed by a General Linear Model, while pollen tube length and pollen area by analysis of variance (ANOVA) and correlation analysis. It was shown that both bioactive products favored in vitro germination of pollen grains cryopreserved for 4 and 8 months, where Biobras-16 at 0.001 mg L-1 showed more consistent results. Pectimorph stimulated pollen tube elongation, which could favor fertilization processes.
