dc.description.abstract | Vaccination with S19 is the best alternative to control bovine brucellosis, but
depending on the immunization protocol, it may induce long-lasting antibodies
interfering with diagnostic tests. Conjunctival vaccinated brucellosis-free heifers
(Farm 1) produced antibodies detected in RBT and cELISA that disappeared <50
days after immunization. In contrast, heifers of Farm 1 subcutaneously vaccinated
with S19 reduced dose produced antibodies detected in 10% of the bovines for up
to one year. Sera of brucellosis-free vaccinated heifers did not immunoprecipitate
native hapten polysaccharide (NH). In contrast, sera of Brucella-infected bovines
readily immunoprecipitated NH. A Brucella abortus infected herd (Farm 2), previously vaccinated with RB51 and subjected to regular tests and slaughter until
reaching negative serology, was mass vaccinated with S19 through either the
conjunctival or subcutaneous routes with reduced and complete doses. Bovines of
Farm 2 displayed sustained and increasing RBT and cELISA positive reactions for
a protracted period, with ~5% bovines reacting against NH, indicating an anamnestic
response upon contact with field Brucella. After an abortion and isolation of field B.
abortus at day 225 in one cow of Farm 2, a significantly higher ratio of cELISA/RBT
reactors was detected (2.5±1.6%) than in preceding days (0.9±0.2). Previous
vaccination with RB51 did not modify the herd brucellosis situation or the serological
results of S19-vaccinated bovines. We conclude that serial testing of RBT-positive
animals with cELISA is not an adequate diagnostic strategy and that the NH
immunoprecipitation test helps identify actively infected cows in herds submitted to
mass vaccination with S19. | es_ES |
dc.description.abstract | La vacunación con S19 es la mejor alternativa para controlar la brucelosis bovina, pero dependiendo del protocolo de inmunización, puede inducir anticuerpos de larga duración que interfieran con las pruebas de diagnóstico. Las novillas libres de brucelosis vacunadas conjuntivalmente (Granja 1) produjeron anticuerpos detectados en RBT y cELISA que desaparecieron <50 días después de la inmunización. En cambio, las novillas de la Granja 1 vacunadas por vía subcutánea con la dosis reducida de S19 produjeron anticuerpos detectados en el 10% de los bovinos hasta un año después. Los sueros de las novillas vacunadas contra la brucelosis no inmunoprecipitaron el hapteno polisacárido nativo (NH). En cambio, los sueros de bovinos infectados por Brucella inmunoprecipitaron fácilmente el NH. Un rebaño infectado por Brucella abortus (Granja 2), previamente vacunado con RB51 y sometido a pruebas periódicas y sacrificio hasta alcanzar serología negativa, fue vacunado masivamente con S19 por vía conjuntival o subcutánea con dosis reducidas y completas. Los bovinos de la granja 2 mostraron reacciones positivas sostenidas y crecientes a RBT y cELISA durante un período prolongado, con ~5% de bovinos reaccionando contra NH, lo que indica una respuesta anamnésica al contacto con Brucella de campo. Tras un aborto y el aislamiento de B. abortus de campo en el día 225 en una vaca de la granja 2, se detectó una proporción significativamente mayor de reactores cELISA/RBT (2,5±1,6%) que en los días precedentes (0,9±0,2). La vacunación previa con RB51 no modificó la situación de la brucelosis en el rebaño ni los resultados serológicos de los bovinos vacunados con S19. Se concluye que las pruebas seriadas de animales positivos a RBT con cELISA no son una estrategia diagnóstica adecuada y que la prueba de inmunoprecipitación de NH ayuda a identificar vacas activamente infectadas en rebaños sometidos a vacunación masiva con S19. | es_ES |
